Most owners have purchased a weekly interval, so shorter stays can be hard to find. If you are willing to pay the weekly price, or if the owner is willing to negotiate, then staying for a shorter interval shouldn't be a problem. You would need to contact the owner directly to make such arrangements. Some owners with points may also be able to reserve just two nights.

I only know Marriott. Yes, one could rent to you for 2 nights. At Marriott, there are membership options, either points or owned weeks. A Marriott owner could make a points reservation for exactly what you want.
If you want a Marriott location, I suggest you disclose here which property is of interest. Someone may then put up a listing for the location/nights you want. Keep in mind that Friday and Saturday nights require 2 or more times the number of points than other nights. Another option would be to place a reservation directly through Marriott, a more costly solution, for either cash or Rewards points.

Marriott, Wyndham, Bluegreen and a few other points systems can reserve shorter stays using points. Problem is that popular locations and times are going to book up early and it isn't likely that someone just happens to be holding onto a reservation for the exact two nights you need. Off season and places with greater availability are more likely to have availability 60 days or less out. Most redweek renters aren't looking to contact and pay for a 2 night stay the 6-10 months out that popular places might require. Also some systems like Wyndham charge a $100 guest certificate fee for each reservation regardless of the length of stay.
Some other weekly timeshares may allow owners to split weeks but this also may require guest certificates and/or additional housekeeping fees that price short stay timeshare much higher add to the fact that check in can sometimes be as late as 5pm and check out can be at 10 am, hotels are often more attractive for one and two night stays.

Holiday Inn Vacation Club point system allows me to rent a two day minimum stay. Booking fee is the same for 2 nights or 2 weeks so a short stay costs a little extra. 10 months in advance , starting and stopping any day of the week. Popular at Christmas / New Years to book 9 or 10 nights to include both holidays at the same place. You do have to plan ahead unless it is off season.
Keep in mind Sunday to Friday (5 nights) stay cost the same as a Friday / Saturday (2 night) stay if your trying to save money.

I am a Club Wyndham Presidential owner and in Club Wyndham we can rent for 2 nights although availability is often an issue. If you have a location, unit size, and dates in mind, I will be happy to check for availability and pricing.
